Chief Chuck Hasty said the bus was destined for New York when it broke down around the South Street area.
The cigarettes, which were packaged and wrapped, were discovered in the storage area of the bus as officers were assisting. The cash was discovered in the possession of a man after officers observed the bag being passed around.
Federal authorities have been notified to assist with the investigation.
Master Officer R. Ball and officers J. Hardy, G. Colson and S. Blythe investigated.
